Deplin® for Depression: How It Can Help
If you've been taking an antidepressant but still aren't feeling like yourself, your healthcare provider may recommend Deplin® (L-methylfolate) as an additional treatment.
Deplin is not an antidepressant, but it can help your antidepressant work more effectively for some people. It contains L-methylfolate, the active form of folate (vitamin B9), which your brain uses to produce important mood-regulating chemicals like serotonin, dopamine, and norepinephrine.
Unlike regular folic acid found in many vitamins, L-methylfolate is already in the form your body can use. This may be especially helpful for people whose bodies have difficulty converting folic acid into its active form.

How Can Deplin Help?
When used along with an antidepressant, Deplin may:
- Help improve symptoms of depression when an antidepressant alone isn't enough
- Increase the effectiveness of your current antidepressant medication
- Support your brain's production of chemicals that regulate mood
- Help some people achieve remission, meaning they experience few or no symptoms of depression
Many people who benefit begin noticing improvement within 4 to 8 weeks, although everyone's experience is different.
Who May Benefit the Most?
While Deplin may help many people with major depressive disorder (MDD), research suggests it may be especially beneficial for individuals who have certain biological factors that can make depression more difficult to treat. These include:
- Being overweight or having obesity (BMI of 30 or higher)
- Higher levels of inflammation in the body, which can sometimes be identified through blood tests
- Low folate (vitamin B9) levels
- Genetic differences, such as an MTHFR gene variant, that make it harder for the body to process folate into the form the brain needs
These factors may affect your brain's ability to produce the neurotransmitters involved in mood. By providing L-methylfolate directly, Deplin helps bypass these barriers and supports healthy neurotransmitter production.
Your healthcare provider may recommend Deplin even if you haven't had genetic testing or blood work, as treatment decisions are based on your overall clinical picture.
What Are the Benefits?
Many patients choose Deplin because it:
- Is generally well tolerated
- Can be taken once daily
- Has very few interactions with other medications
- Does not typically cause weight gain, sexual side effects, or sedation
- Can be safely combined with most antidepressants
Is It Right for Everyone?
Not everyone with depression will benefit from Deplin, and it is not intended to replace your antidepressant medication. Instead, it is used as an "add-on" treatment to help improve your response.
Your healthcare provider will consider your symptoms, medical history, and current medications to determine whether Deplin may be a good option for you.
Talk With Your Provider
If your depression symptoms have improved only partially—or not as much as you'd hoped—talk with your provider about whether Deplin may be an appropriate addition to your treatment plan. Together, you can decide if this evidence-based option is right for you.
